Osmanlı da Ayasofya Camisi adıyla anmış.Kıbrıs II. Sultan Selim’in döneminde fethedildiğinden, caminin adı Kıbrıs Müftüsü’nün 13.8.1954 tarihli emriyle ‘’Selimiye Camisi’’ olarak değiştirilmiş.

Osmanlı Dönemi boyunca sadece Lefkoşa’nın değil tüm adanın da en büyük camisi olarak hizmet vermiş. Osmanlı valileri, üst düzey yöneticiler ve kent ileri gelenleri Cuma namazını bu camide kılarlarmış.
